Both he and his father, TDP chief, Chandrababu Naidu suffered a major political setback in the 2019 Andhra Pradesh Legislative Assembly Elections by winning only 23 seats out of the total 175 seats. This was TDP's worst defeat ever in the history.

Following the defeat, Lokesh embarked on a 3,100km padayatra, covering 100 assembly constituencies over 226 days. Despite facing legal challenges during this period, including the Inner Ring Road (IRR) case, 6 Lokesh managed to enhance his political standing and emerged as a prominent leader within the TDP.
